Behavior effect
Social behaviour and mood in everyday life: the effects of tryptophan in quarrelsome individuals.

We hypothesized that increasing brain serotonin in healthy individuals with high scores on 2 self-report measures of trait quarrelsomeness would reduce quarrelsome behaviours and enhance agreeable behaviours when measured ecologically using an event-contingent recording method. We conducted a double-blind crossover study, in which participants took l-tryptophan at 3 grams a day and placebo for 15 days each and recorded how they behaved, felt and perceived others during everyday social interactions. L-Tryptophan supplement use significantly decreased quarrelsome behaviours and increased agreeable behaviours and perceptions of agreeableness. Men also behaved less dominantly, whereas both men and women perceived others as more dominant. L-Tryptophan's effects on behaviours and perceptions, while more marked in the men, were generally positive and accompanied by improved affect. Increasing serotonin in quarrelsome people may not only reduce behaviours associated with a predisposition to various mental and physical disorders but also enhance socially constructive behaviours and improve social perceptions.

Sleep and insomnia
Dietary carbohydrate intake has been shown to increase the plasma concentration of l-tryptophan, a precursor of serotonin and sleep-inducing agent.

Does this really work for insomnia? Wasn't this amino acid banned just a few years ago? I assume it is now legal or you would not be selling it. If I took this product should I take something in addition to it? I have had insomnia for years, tried so many products and nothing seems to work. I try to stay away from prescription drugs like Ambien but sometimes I find that is the only thing that "knocks" me out.
    Yes, it is now legal and some people find it helpful for insomnia although those who are used to taking potent prescription sleeping pills may not find it strong enough.

Intact protein rich in tryptophan was not seen as an alternative to pharmaceutical grade tryptophan since protein also contains large neutral amino acids (LNAAs) that compete for transport sites across the blood-brain barrier (BBB). Deoiled gourd seed (an extremely rich source of tryptophan - 22 mg tryptophan /1 g protein) was combined with glucose, a carbohydrate that reduces serum levels of competing LNAAs which was then compared to pharmaceutical grade tryptophan with carbohydrate as well as carbohydrate alone. Subjects were randomly assigned to one of three conditions: (1) Protein source tryptophan (deoiled gourd seed) in combination with carbohydrate; (2) pharmaceutical grade tryptophan in combination with carbohydrate; (3) carbohydrate alone. Protein source tryptophan with carbohydrate and pharmaceutical grade tryptophan, but not carbohydrate alone, resulted in significant improvement on subjective and objective measures of insomnia. Metabolism and biochemistry
L-Tryptophan converts into Hydroxy tryptophan, which then readily converts into serotonin which has a brain and mood influence, there are several nutrients that increase serotonin levels. Once serotonin is made, the pineal gland is able to convert it at night into melatonin, the sleep-inducing hormone.

Pregnancy
I am very interested to use 5 HTP and Tryptophan supplements. My question is if can I use both products if I am pregnant? I actually take Prozac because I have a big problem with serotonin.
   We are not aware of any human studies in women who are pregnant who have been placed on long term 5-HTP or tryptophan supplement treatment. Since we don't know if these supplements are safe during pregnancy, we cannot advise them to be used.

Interactions with medications
Is it safe to take tryptophan with Tofranil?
    Tofranil, imipramine hydrochloride USP, the original tricyclic antidepressant, is a member of the dibenzazepine group of compounds. It is not easy to predict interactions between medications and supplements and dosage has a major factor.

The history
During the 1980s consumers were using l-tryptophan for sleep and as an antidepressant. L-Tryptophan was available without prescription until 1989 when the FDA prohibited its over-the-counter sale because a l-tryptophan manufacturer in Japan shipped a contaminated batch to the U.S. This caused a serious illness called eosinophilia myalgia syndrome in about 1,000 individuals.
Around 1995, l-tryptophan gradually became available by prescription through compounding pharmacies, and then since about the year 2000 l-tryptophan slowly and cautiously was placed on the over the counter market through a few small vitamin companies. Few large vitamin companies are selling l-tryptophan at this time (2008).

I am 98 Kg and had no food 3 hours before taking it. I took 4 tryptophan capsules with water and was still awake after 3 hours. I took 2 more capsules still could not sleep. One hour later I took 2 more capsules and have been awake all night and could not sleep. I took a total of 8 capsules and the L-Tryptophan and it had no effect on me. Does it take a while to get into your system.
   Sometimes taking too much of a certain herb or supplement can cause stimulation as opposed to sedation. It is better to stay with 500 mg of l-tryptophan a few hours before bed. A better option for sleep is Good Night Rx which is much more consistent.

Q. I was just wondering why the directions on a l-tryptophan web site say to take only 4 days a week. I work nights, and have a horrible time sleeping, expecially when I am off.
   A. Tolerance can develop to most pills used for sleeping, therefore it is better not to use them every night so that their benefit continues longer. Some people can take Good Night Rx 2 or 3 times a week as a substitute for the times a l-tryptophan supplement is not taken. However, it is still best not to take a pill every night of the week.

Q. I currently take a tryptophan supplement at night but would like to try taking part of my dosage in the morning to see if more of it might convert to serotonin. I have a thyroid problem. I am hypothyroid and take 112mcg of Synthroid each morning. Would it be ok to take both the Synthroid and tryptophan in the morning before eating?
   A. This is a question for your doctor since different people may respond differently. When adding a new supplement, it is often a good idea to first use a low dosage even if it requires taking a portion of a capsule.

Q. Does Taking 4000 mg of Tryptophan per day( overdosing) for a long period of time harmful? A doctor told me that it would cause stiffness in the tissues and even bleeding internally. I was wondering would it be harmful to take 500 mg of tryptophan per day for a period of 1 year? Also how much of tryptophan gets converted to serotonin comparing to 5-htp?
   A. Tryptophan converts to 5-HTP which converts to serotonin. Therefore, much less 5-HTP is needed to convert to serotonin that tryptophan. The exact percentage of conversion of tryptophan to serotonin varies among individuals and it can vary depending on time of day and whether there is food in the stomach or not. There are many variables that influence the conversion of tryptophan to 5htp to serotonin so there is no simple answer. Plus, different tissues and organs of the body may have a different conversion rate.
   Long term human studies with high dose tryptophan supplements are not available to know the safety and danger or long term tryptophan side effects. It is a good idea, until we learn more, to limit the use of tryptophan supplements to 5 days a week with a week off each month. This is just a rough guideline.

I'm curious whether I can take the three supplements I am ordering at the same time...Tryptophan, Passion Rx and Resveratrol. I'm 60, take only vitamins and have no medical condition. Plan to take only low dosage.
    We suggest taking them separately for a few days before considering combining them.

Can 5 HTP boost levels of norepenephrine: what other supplements do? Are L tryptophan and 5HTP ever taken together? What's the difference? From what I've read & learned it seems, L tryptophan is more connected with serotonin, than noreprenephrin.
    5-HTP has more of an effect on serotonin levels, its actions are similar to and overlap with tryptophan, if both are taken together the dosages need to be reduced. Tyrosine and phenylalanine are more likely to effect norepinephrine levels.

I have read that when taken in the morning, tryptophan will be converted to seratonin, but when taken at night, tryptophan will be converted to melatonin. Therefore, if tryptophan is being taken for depression, would taking it in the morning rather than the night be advisable?
   Before converting to melatonin, tryptophan converts into serotonin. Most people find mood elevating properties of this amino acid whether taken during the day or evening but most people find it to be effective taken in the evening.

Thank you for your informative website, I am interested in some of natural products but wanted to ask a question about an interaction I had had with a different product because I want to avoid it happening again if I try something else. After doing some research I decided to take 500mg of tryptophan (Jarrow brand I think) to begin with before bed, doubling up to 1g after a couple of nights. It worked well at first, however after a few nights as I was drifting off to sleep I felt an electrical buzzing sensation at the front of my head, which came in waves and kept me awake all night. Needless to say I stopped taking this but it has left me wondering whether it was a poor quality product or whether this was a unique interaction with my chemistry. I appreciate you can't offer medical advice on my individual
circumstances but I wonder if you could comment generally on this type of side effect and whether their are other types of amino acids I should avoid.
    Sometimes taking too high a dosage of a supplement can cause unexpected reactions, and some people have reported this kind of strange sensation with 5-htp or tryptophan.

I am currently taking tryptophan, gaba, sedaplus and melatonin to help me sleep. Also one of my other supplements is methionine. I was wondering if any of those could help cause a amaurosis fugax. Had it to happen last night. Couldn't really see out of one eye.... very dark gray... lasted a few minutes. My eye felt strange as well. Went to optometrist and things look ok. I saw something that a couple of these supplements could cause vasospasms even in the eye. Have you all heard or know of such research? A little afraid to continue with supplements.
